Total RT volume is 100microL containing 10mM Tris-HCl (pH 8.3), 50mM KCl, 1.5mM MgCl2, 0.01% gelatine, 10mM DTT, 100mM dNTPs, 0.4U/microL human placenta ribonuclease inhibitor, 2ng/microL primer 3D2, and 0.1U/microL AMV RTase (Seikagaku America).

After 42C for 40min, PCR is performed by adding 2ng/microL primer 3D-1 and polymerase to the reaction.
